This variant lies in the MYLK gene (transcript NM_053025.4) at coding-DNA position 250, where G is replaced by A; at the protein level this means replaces glycine at residue 84 with serine — a missense variant. Submitter rationale: Variant summary: MYLK c.250G>A (p.Gly84Ser) results in a non-conservative amino acid change located in the Immunoglobulin subtype 2 domain (IPR003598) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 251188 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.250G>A in individuals affected with Thoracic Aortic Aneurysms And Dissections and no experimental evidence demonstrating its impact on protein function have been reported.
