Submitter rationale: Variant summary: BRCA2 c.5343_5344insA (p.Gln1782ThrfsX5) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 250696 control chromosomes (gnomAD). Variants which result in similar protein level effect have been reported in the literature in individual(s) affected with Hereditary Breast and Ovarian Cancer Syndrome (e.g. de Juan Jimenez_2013). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
