Uncertain significance for Cardiomyopathy — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_004415.4(DSP):c.314G>A (p.Arg105Gln), citing ACMG Guidelines, 2015: This missense variant replaces arginine with glutamine at codon 105 of the DSP protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with sudden unexplained death (PMID: 25447171) and in another individual affected with ischemic heart failure (doi:10.3390/cardiogenetics12020018). This variant has been identified in 6/251478 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
