Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000251.3(MSH2):c.2105T>G (p.Val702Gly), citing Ambry Variant Classification Scheme 2023: The p.V702G variant (also known as c.2105T>G), located in coding exon 13 of the MSH2 gene, results from a T to G substitution at nucleotide position 2105. The valine at codon 702 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
