NM_000492.4(CFTR):c.8G>T (p.Arg3Met) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3M variant (also known as c.8G>T), located in coding exon 1 of the CFTR gene, results from a G to T substitution at nucleotide position 8. The arginine at codon 3 is replaced by methionine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
