Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_014244.5(ADAMTS2):c.1349G>A (p.Arg450His), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ADAMTS2 c.1349G>A (p.Arg450His) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 5.2e-05 in 230176 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in ADAMTS2 causing Ehlers-Danlos syndrome, dermatosparaxis type (5.2e-05 vs 0.0011), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1349G>A in individuals affected with Ehlers-Danlos syndrome, dermatosparaxis type and no experimental evidence demonstrating its impact on protein function have been reported.
