This variant lies in the ADAMTS2 gene (transcript NM_014244.5) at coding-DNA position 2842, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 948 with asparagine — a missense variant. Submitter rationale: Variant summary: ADAMTS2 c.2842G>A (p.Asp948Asn) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 4.1e-06 in 245952 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2842G>A in individuals affected with Ehlers-Danlos Syndrome, Type VIIC (Dermatosparaxis) and no experimental evidence demonstrating its impact on protein function have been reported. Two submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. Both submitters classified the variant as uncertain significance.
