Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_014244.5(ADAMTS2):c.2930G>A (p.Gly977Asp), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the ADAMTS2 gene (transcript NM_014244.5) at coding-DNA position 2930, where G is replaced by A; at the protein level this means replaces glycine at residue 977 with aspartic acid — a missense variant. Submitter rationale: Variant summary: ADAMTS2 c.2930G>A (p.Gly977Asp) results in a non-conservative amino acid change located in the Thrombospondin type 1 repeats (IPR000884) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 248246 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.2930G>A in individuals affected with Ehlers-Danlos syndrome, dermatosparaxis type and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 905621). Based on the evidence outlined above, the variant was classified as uncertain significance.
