Uncertain significance for MEGF10-related myopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001256545.2(MEGF10):c.1906G>A (p.Gly636Arg), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with MEGF10-related conditions. ClinVar contains an entry for this variant (Variation ID: 904940). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant is present in population databases (rs201512679, ExAC 0.1%). This sequence change replaces glycine with arginine at codon 636 of the MEGF10 protein (p.Gly636Arg). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine.
