Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.838T>G (p.Tyr280Asp), citing Ambry Variant Classification Scheme 2023: The p.Y280D variant (also known as c.838T>G), located in coding exon 10 of the MLH1 gene, results from a T to G substitution at nucleotide position 838. The tyrosine at codon 280 is replaced by aspartic acid, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
