Pathogenic for Atrioventricular septal defect 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001308093.3(GATA4):c.889G>A (p.Gly297Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 296 of the GATA4 protein (p.Gly296Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with congenital heart defects (PMID: 12845333). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 9030). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GATA4 protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects GATA4 function (PMID: 12845333). For these reasons, this variant has been classified as Pathogenic.
