Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.8375C>G (p.Ser2792Cys), citing Ambry Variant Classification Scheme 2023: The p.S2792C variant (also known as c.8375C>G), located in coding exon 26 of the APOB gene, results from a C to G substitution at nucleotide position 8375. The serine at codon 2792 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
