NM_000104.4(CYP1B1):c.850C>T (p.Arg284Trp) was classified as Uncertain significance for Congenital glaucoma by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with tryptophan at codon 284 of the CYP1B1 protein (p.Arg284Trp). The arginine residue is weakly conserved and there is a moderate physicochemical difference between arginine and tryptophan. This variant is present in population databases (rs368249322, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with CYP1B1-related conditions. ClinVar contains an entry for this variant (Variation ID: 895299). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CYP1B1 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
