Uncertain significance for Malignant hyperthermia, susceptibility to, 1 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000540.3(RYR1):c.11119C>T (p.Arg3707Cys), citing ACMG Guidelines, 2015: This missense variant replaces arginine with cysteine at codon 3707 of the RYR1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR1-related disorders in the literature. This variant has been identified in 3/248688 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The same amino acid change resulting from a different DNA substitution (c.11120G>T) has been observed in an individual with malignant hyperthermia susceptibility (PMID: 16917943). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
