Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001384474.1(LOXHD1):c.5891G>T (p.Gly1964Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the LOXHD1 gene (transcript NM_001384474.1) at coding-DNA position 5891, where G is replaced by T; at the protein level this means replaces glycine at residue 1964 with valine — a missense variant. Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1902 of the LOXHD1 protein (p.Gly1902Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal recessive deafness (Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 892535). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
