Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001143992.2(WRAP53):c.1537C>G (p.Arg513Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with glycine, which is neutral and non-polar, at codon 513 of the WRAP53 protein (p.Arg513Gly). This variant is present in population databases (rs144549494, gnomAD 0.1%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with WRAP53-related conditions. ClinVar contains an entry for this variant (Variation ID: 891911). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
