Uncertain significance for T-cell immunodeficiency, congenital alopecia, and nail dystrophy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001369369.1(FOXN1):c.1727C>T (p.Pro576Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 576 of the FOXN1 protein (p.Pro576Leu). This variant is present in population databases (rs756165992, gnomAD 0.04%). This variant has not been reported in the literature in individuals affected with FOXN1-related conditions. ClinVar contains an entry for this variant (Variation ID: 888888). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
