Uncertain significance for Achondrogenesis, type IA — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004239.4(TRIP11):c.3743A>G (p.Gln1248Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with arginine at codon 1248 of the TRIP11 protein (p.Gln1248Arg). The glutamine residue is weakly conserved and there is a small physicochemical difference between glutamine and arginine. This variant is present in population databases (rs150694723, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with TRIP11-related conditions. ClinVar contains an entry for this variant (Variation ID: 886443). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TRIP11 protein function.
