Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000260.4(MYO7A):c.5018T>A (p.Val1673Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine with aspartic acid at codon 1673 of the MYO7A protein (p.Val1673Asp). The valine residue is moderately conserved and there is a large physicochemical difference between valine and aspartic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with MYO7A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15").
