Uncertain significance for Familial hemophagocytic lymphohistiocytosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001083116.3(PRF1):c.1111A>G (p.Arg371Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glycine at codon 371 of the PRF1 protein (p.Arg371Gly). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and glycine. This variant is present in population databases (rs566891566, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with PRF1-related conditions. ClinVar contains an entry for this variant (Variation ID: 878087). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
