Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001002294.3(FMO3):c.1322T>C (p.Ile441Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 441 of the FMO3 protein (p.Ile441Thr). This variant is present in population databases (rs535435928, gnomAD 0.03%). This missense change has been observed in individual(s) with trimethylaminuria (PMID: 22819296, 30351217, 35853340). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 875899). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on FMO3 protein function. Experimental studies have shown that this missense change affects FMO3 function (PMID: 22819296, 30351217). For these reasons, this variant has been classified as Pathogenic.
