NM_013339.4(ALG6):c.370T>G (p.Tyr124Asp) was classified as Uncertain significance for ALG6-congenital disorder of glycosylation 1C by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 124 of the ALG6 protein (p.Tyr124Asp). This variant is present in population databases (rs562509297, gnomAD 0.08%), including at least one homozygous and/or hemizygous individual. This variant has not been reported in the literature in individuals affected with ALG6-related conditions. ClinVar contains an entry for this variant (Variation ID: 875420). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
