Likely pathogenic for Polycystic kidney disease, adult type — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001009944.3(PKD1):c.1606+5G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: PKD1 c.1606+5G>A alters a non-conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 5' splicing donor site. Two predict the variant weakens a 5' donor site. Two predict the variant creates a cryptic 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 135224 control chromosomes. c.1606+5G>A has been observed in the heterozygous state in multiple individuals affected with Polycystic Kidney Disease 1 (e.g., Ercoskun_2025, Benson_2021, Zhang_2019). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
