Uncertain significance for Amyotrophic lateral sclerosis type 10; FRONTOTEMPORAL LOBAR DEGENERATION WITH TDP43 INCLUSIONS, TARDBP-RELATED — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_007375.4(TARDBP):c.893G>T (p.Gly298Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 298 of the TARDBP protein (p.Gly298Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with amyotrophic lateral sclerosis (PMID: 32579787, 35239007; Invitae). ClinVar contains an entry for this variant (Variation ID: 873205). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant disrupts the p.Gly298 amino acid residue in TARDBP. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 18396105, 20558945, 28709720, 32166880). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
