Likely pathogenic — the classification assigned by GeneDx to NM_004699.4(FAM50A):c.763G>A (p.Asp255Asn), citing GeneDx Variant Classification (06012015): The D255N variant in the FAM50A gene has been observed as a de novo hemizygous variant in GeneDx whole exome sequencing data in association with global developmental delay, short stature, small hands and feet, dysmorphic features, ocular anomalies, and seizures. The D255N variant is not observed in large population cohorts (Lek et al., 2016). In silico analysis supports that this missense variant has a deleterious effect on protein structure/function. Therefore, we interpret D255N as a likely pathogenic variant.
