This variant lies in the ATP13A2 gene (transcript NM_022089.4) at coding-DNA position 1847, where A is replaced by C; at the protein level this means replaces glutamic acid at residue 616 with alanine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid with alanine at codon 616 of the ATP13A2 protein (p.Glu616Ala). The glutamic acid residue is weakly conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant is present in population databases (rs558165485, ExAC 0.05%). This variant has not been reported in the literature in individuals affected with ATP13A2-related conditions.
