Pathogenic for Autosomal recessive limb-girdle muscular dystrophy — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000232.5(SGCB):c.323T>G (p.Leu108Arg), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the SGCB gene (transcript NM_000232.5) at coding-DNA position 323, where T is replaced by G; at the protein level this means replaces leucine at residue 108 with arginine — a missense variant. Submitter rationale: Variant summary: SGCB c.323T>G (p.Leu108Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251476 control chromosomes. c.323T>G has been observed in multiple individuals affected with Autosomal Recessive Limb-Girdle Muscular Dystrophy (e.g. Moreira_2003, Alcantara-Ortigoza_2019, Bevilacqua_2024). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 31671740, 39678382, 8968749, 12566530, 8968750). ClinVar contains an entry for this variant (Variation ID: 8717). Based on the evidence outlined above, the variant was classified as pathogenic.
