Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 362 of the RUNX2 protein (p.Ala362Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with cleidocranial dysplasia (PMID: 12196916; internal data). In at least one individual the variant was observed to be de novo. This variant is also known as c.1043C>T, A348V. ClinVar contains an entry for this variant (Variation ID: 870903). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
