Uncertain significance for Catecholaminergic polymorphic ventricular tachycardia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006073.4(TRDN):c.425-4_425-3insLINE1, citing Invitae Variant Classification Sherloc (09022015): This sequence change inserts a L1 transposable element in intron 4 of the TRDN mRNA (c.425-4_425-3insLINE1). The exact size and sequence of the insertion cannot be determined by the current assay. However, the insertion is expected to result in an absent or disrupted protein product. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with TRDN-related disease. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site, but this prediction has not been confirmed by published transcriptional studies.
