This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 5174, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 1725 with glycine — a missense variant. Submitter rationale: The p.E1725G variant (also known as c.5174A>G), located in coding exon 17 of the BRCA1 gene, results from an A to G substitution at nucleotide position 5174. The glutamic acid at codon 1725 is replaced by glycine, an amino acid with similar properties. One functional study found that this nucleotide substitution is functional in a high throughput genome editing haploid cell survival assay (Findlay GM et al. Nature, 2018 10;562:217-222).
