This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 5563, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1855 with valine — a missense variant. Submitter rationale: This missense variant replaces isoleucine with valine at codon 1855 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). A functional study has reported that this variant does not impact BRCA1 function in a haploid cell proliferation assay (PMID: 30209399). This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
