Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_007294.4(BRCA1):c.5417C>T (p.Pro1806Leu), citing ACMG Guidelines, 2015: This missense variant replaces proline with leucine at codon 1806 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function. Functional studies have reported that this variant does not impact BRCA1 in a haploid cell proliferation assay and a homology-mediated DNA repair assay (PMID: 30209399, 35196514). To our knowledge, this variant has not been reported in individuals affected with BRCA1-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
