Submitter rationale: To the best of our knowledge, the BRCA1 c.23T>C (p.V8A) variant has not been reported in individuals with BRCA1-related disease. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org), but has been reported in ClinVar (Variation ID: 867693). This variant involves a moderately conserved amino acid, and computational analyses suggest that the variant does not impact the function of the protein. However, these predictions have not been confirmed by published functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
