Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000539.3(RHO):c.1045T>C (p.Ter349Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change disrupts the translational stop signal of the RHO mRNA. It is expected to extend the length of the RHO protein by 51 additional amino acid residues. This variant is not present in population databases (gnomAD no frequency). This protein extension has been observed in individuals with autosomal dominant retinitis pigmentosa (PMID: 16799052, 31960602; internal data). This variant is also known as Ter345Gln. ClinVar contains an entry for this variant (Variation ID: 866186). This variant results in an extension of the RHO protein. Other variant(s) that result in a similarly extended protein product (p.*349Glnext*51) have been determined to be pathogenic (PMID: 10189219, 23940033). This suggests that these extensions are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
