This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 237, where T is replaced by A; at the protein level this means replaces phenylalanine at residue 79 with leucine — a missense variant. Submitter rationale: Variant summary: BRCA1 c.237T>A (p.Phe79Leu) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251192 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.237T>A in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome has been reported. At least one publication reports experimental evidence evaluating an impact on protein function (Findlay_2018). These results showed no damaging effect of this variant on homology directed repair activity in-vitro (HDR). No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
