Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.529G>C (p.Glu177Gln), citing Ambry Variant Classification Scheme 2023: The p.E177Q variant (also known as c.529G>C), located in coding exon 1 of the AXIN2 gene, results from a G to C substitution at nucleotide position 529. The glutamic acid at codon 177 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
