Uncertain significance for Autoimmune lymphoproliferative syndrome, type III caused by mutation in PRKCD — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006254.4(PRKCD):c.1211C>G (p.Ala404Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with glycine at codon 404 of the PRKCD protein (p.Ala404Gly). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs782150167, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with PRKCD-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C55"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
