NM_000051.4(ATM):c.1802G>A (p.Ser601Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1802G>A variant (also known as p.S601N), located in coding exon 10 of the ATM gene, results from a G to A substitution at nucleotide position 1802. The amino acid change results in serine to asparagine at codon 601, an amino acid with highly similar properties. However, this change occurs in the last base pair of coding exon 10, which makes it likely to have some effect on normal mRNA splicing. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico protein analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
