Pathogenic for Emery-Dreifuss muscular dystrophy 4, autosomal dominant; Autosomal recessive ataxia, Beauce type — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_182961.4(SYNE1):c.2536C>T (p.Gln846Ter), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Gln853*) in the SYNE1 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in SYNE1 are known to be pathogenic (PMID: 27086870). This variant has not been reported in the literature in individuals with SYNE1-related conditions. This variant is not present in population databases (ExAC no frequency).
