This variant lies in the CLCN1 gene (transcript NM_000083.3) at coding-DNA position 1247, where G is replaced by A; at the protein level this means replaces glycine at residue 416 with glutamic acid — a missense variant. Submitter rationale: Variant summary: CLCN1 c.1247G>A (p.Gly416Glu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251470 control chromosomes. c.1247G>A has been reported in the literature in at-least two unrelated individuals affected with Myotonia congenita (Mazon_2012, Milla_2019). These data indicate that the variant may be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in leak and capacitive currents in HEK cells (Mazon_2012).
