Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014168.4(METTL5):c.380C>G (p.Pro127Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces proline with arginine at codon 127 of the METTL5 protein (p.Pro127Arg). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and arginine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with METTL5-related intellectual disability (Invitae). It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
