Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_005802.5(TOPORS):c.2422G>T (p.Glu808Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TOPORS gene (transcript NM_005802.5) at coding-DNA position 2422, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 808 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change creates a premature translational stop signal (p.Glu808*) in the TOPORS gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 238 amino acid(s) of the TOPORS protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal dominant retinitis pigmentosa (PMID: 18509552). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 861205). This variant is located in a region of the TOPORS protein where a significant number of TOPORS nonsense and frameshift mutations have been reported in association with autosomal dominant retinitis pigmentosa (PMID: 35579903, 17924349, 32531858). For these reasons, this variant has been classified as Pathogenic.
