Uncertain significance for Primary familial hypertrophic cardiomyopathy; Dilated cardiomyopathy 1AA — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001103.4(ACTN2):c.698-10_1107+10del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 8-10 of the ACTN2 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. This variant has not been reported in the literature in individuals with ACTN2-related conditions. The current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in ACTN2 cause disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
