Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_024529.5(CDC73):c.965C>G (p.Ser322Cys), citing Ambry Variant Classification Scheme 2023: The p.S322C variant (also known as c.965C>G), located in coding exon 10 of the CDC73 gene, results from a C to G substitution at nucleotide position 965. The serine at codon 322 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
