Pathogenic for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000251.3(MSH2):c.955_959del (p.Asp319fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 955 through coding-DNA position 959, deleting 5 bases; at the protein level this means shifts the reading frame starting at aspartic acid residue 319, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Asp319Hisfs*12) in the MSH2 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in MSH2 are known to be pathogenic (PMID: 15849733, 24362816). This variant has not been reported in the literature in individuals with MSH2-related conditions. This variant is not present in population databases (ExAC no frequency).
