The arginine at codon 2123 is replaced by histidine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with dilated cardiomyopathy (DCM) (Janin A et al. Mol Diagn Ther, 2021 May;25:373-385). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
