This variant lies in the MLH1 gene (transcript NM_000249.4) at coding-DNA position 1558, where G is replaced by A; at the protein level this means replaces valine at residue 520 with isoleucine — a missense variant. Submitter rationale: This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Nucleotide substitutions within the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098).
