Uncertain Significance — the classification assigned by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories to NM_000038.6(APC):c.2932C>A (p.Gln978Lys), citing ARUP Molecular Germline Variant Investigation Process 2024. This variant lies in the APC gene (transcript NM_000038.6) at coding-DNA position 2932, where C is replaced by A; at the protein level this means replaces glutamine at residue 978 with lysine — a missense variant. Submitter rationale: The APC c.2932C>A; p.Gln978Lys variant (rs1765298309), to our knowledge, is not reported in the medical literature but is reported in ClinVar (Variation ID: 859020). This variant is also absent from the Genome Aggregation Database (v2.1.1), indicating it is not a common polymorphism. Computational analyses are uncertain whether this variant is neutral or deleterious (REVEL: 0.496). The vast majority of pathogenic APC variants are truncating nonsense or frameshift variants (see InSiGHt, Kerr 2013). However, given the lack of clinical and functional data, the significance of this variant is uncertain at this time.
