This variant lies in the CACNB4 gene (transcript NM_000726.5) at coding-DNA position 495, where A is replaced by T; at the protein level this means replaces glutamic acid at residue 165 with aspartic acid — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
