Pathogenic for Familial hypokalemia-hypomagnesemia — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001126108.2(SLC12A3):c.1261T>C (p.Cys421Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: SLC12A3 c.1261T>C (p.Cys421Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.1261T>C has been observed in individual(s) affected with Gitelman syndrome (example: Simon_1996, internal data). These data indicate that the variant is likely to be associated with disease. A different variant affecting the same codon has been classified as likely pathogenic by our lab (c.1262G>A,p.Cys421Tyr), supporting the critical relevance of codon 421 to SLC12A3 protein function.
